Peptide Storage: Temperature, Humidity & Shelf-Life Explained

How you store lyophilized peptides directly determines how long they remain suitable for research. This guide covers temperature recommendations, humidity control, shelf-life timelines, and storage best practices based on current analytical chemistry literature.

Optimal temperature for lyophilized peptides

The gold-standard storage temperature for most research-grade lyophilized peptides is –20 °C (a standard laboratory freezer). At this temperature, lyophilized peptides typically remain stable and usable for 12–36 months, depending on the specific peptide and storage conditions.

For extended stability beyond 3 years, –80 °C (an ultra-low freezer) is preferred. Studies on freeze-dried preparations (including peptide vaccines) have shown full stability at –80 °C for five years or more with no measurable degradation.

Humidity: the critical threat

Humidity is the single most damaging factor for lyophilized peptides. Peptides are highly hygroscopic — they absorb moisture from the air readily. Even modest rehydration can trigger:

Best practice: Store lyophilized peptides in airtight, light-resistant vials with desiccant packs (silica gel or molecular sieves). Avoid repeated opening; use only what you need and reseal immediately.

Freeze-thaw cycles and degradation

Repeated freeze-thaw cycles damage peptides by causing ice-crystal formation that disrupts the lyophilized matrix. Limit freeze-thaw to 3–4 cycles maximum. Strategy: aliquot reconstituted peptide into small volumes before freezing, so you thaw only what you use per experiment.

Light sensitivity

Some peptides, especially those containing aromatic amino acids (tryptophan, tyrosine), are light-sensitive. Photodegradation can occur even in dark freezers over months. Use amber or opaque vials whenever possible, and store in light-blocked containers.
